Analysis of the possible association of patients’ genetic factors with course of dengue virus infection

Abstract

The aim of the study was to investigate association of genes polymorphisms of the innate immunity and blood clotting system with different courses of dengue fever (DF).

Material and methods. The following groups of subjects were formed: dengue hemorrhagic fever (DHF) (n=13), classic DF (n=63), and a comparison group (blood donor candidates, n=62). The subjects’ DNA from venous blood was analyzed. The certified kits “ImmunoGenetics IL28B” and “CardioGenetics Thrombophilia” (DNA Technology, Russian Federation) were used to identify allelic variants at polymorphic loci. The known polymorphisms of the genes IFNL3, IFNL4, FGB, F2, F5, F7, F13, ITGA2, ITGB3, and PAI-1 were analyzed.

Results. As a result of this comparative study, no association was found between single nucleotide polymorphism (SNP) in the IFNL3 (rs8099917 T>G) and IFNL4 (rs12979860 C>T) genes and the DF course. The analysis of polymorphism of blood clotting genes (Factors I, II, V, VII, XIII), collagen and fibrinogen platelet receptor genes, and tissue plasminogen activator antagonist gene has shown that the A allele in F7 gene (SNP 10976 G>A) was detected more often in patients with DHF and classic DF than in the comparison group. The presence of the A allele, which is known to reduce blood clotting and increase hemorrhage, probably complicates the pathogenesis of DF. The presence of many unfavorable alleles (7 of 10 analyzed genes) was revealed in one patient with a lethal outcome.

Conclusion. The polymorphisms of IFNL3 and IFNL4 genes, that encode and affect the expression of IFN-λ3, did not affect the courses of DF. Among all analyzed blood coagulation genes, an association between the presence of the A allele, that reduces blood clotting, and DF/DHF was found in the F7 gene (SNP 10976 G>A) only. This data provides evidence of a genetic predisposition to a severe course of DF.
